1、第36卷第lO期2013年lO月北京中醫(yī)藥大學(xué)學(xué)報(bào)Vo1.36No.10Oct.2013JournalofBeijingUniversityofTraditionalChineseMedicine705多葉棘豆總黃酮大孔吸附樹(shù)脂純化工藝鐘昆芮孫芳芳折改梅劉斌(北京中醫(yī)藥大學(xué)中藥學(xué)院北京100102)摘要:目的研究大孔吸附樹(shù)脂純化多葉棘豆總黃酮的工藝條件。方法以總黃酮含量和吸附一解吸率為指標(biāo),考察5種不同大孔吸附樹(shù)脂對(duì)總黃酮的吸附能力,確定D4006型樹(shù)脂為純化用樹(shù)脂,對(duì)純化工藝中吸附條件、除雜條件和洗脫條件進(jìn)行優(yōu)化。結(jié)果最佳工藝條件為上樣液濃
2、度0.05g/mL,最大上樣量24倍柱體積(BV),吸附流速3BV/h,樹(shù)脂柱徑高比1:9,待吸附結(jié)束后,用5%乙醇洗脫9BV進(jìn)行除雜,再用50%乙醇以3BV/h的流速洗脫8BV,經(jīng)IMO06型大孔吸附樹(shù)脂純化后,多葉棘豆總黃酮含量可達(dá)54%。結(jié)論所建立的工藝分離效果良好,操作簡(jiǎn)單,重復(fù)性好,可用于分離富集多葉棘豆總黃酮。關(guān)鍵詞:多葉棘豆;總黃酮;大孔吸附樹(shù)脂;純化中圖分類號(hào):R284.2doi:10.3969/j.issn.1006.2157.2013.10.013Purificationtechnologyoftotalflavonoid
