1、大孔吸附樹脂純化刺玫果總黃酮工藝研究論文摘要刺玫果學(xué)名野薔薇,又名野刺玫果,山刺玫果,系薔薇科薔薇屬植物山刺玫的成熟果實(shí)。廣泛分布于我國(guó)的東北地區(qū),尤以黑龍江省貯量最為豐富?,F(xiàn)代藥理學(xué)研究表明:刺玫果含有豐富的營(yíng)養(yǎng)成分及生物活性物質(zhì),具有健脾理氣、養(yǎng)血調(diào)經(jīng)、抗衰老、防治心血管疾病、增強(qiáng)免疫及促智等功效。刺玫果中所含的總黃酮類物質(zhì)具有預(yù)防和治療高血脂、血栓所致的心腦血管疾病的功效,且具有高效、低毒副作用的優(yōu)勢(shì)。因此純化刺玫果中的總黃酮就具有現(xiàn)實(shí)的科研意義。目前,常見的總黃酮純化方法有水提醇沉法(水醇
2、法)、醇提水沉法(醇水法)、酸堿法、鹽析法、離子交換法和結(jié)晶法等。實(shí)踐表明,這些方法也存在一定的局限性。近年出現(xiàn)了一些分離和純化的新方法,如絮凝沉淀法,大孔樹脂吸附法、超濾法、高速離心法等。本論文采用大孔樹脂法純化刺玫果提取物中的總黃酮。探索洗脫劑、pH值、上樣濃度等因素對(duì)純化過程的影響。采用紫外分光光度計(jì)法對(duì)總黃酮含量進(jìn)行了分析,最終確定了洗脫劑的濃度75%、上樣PH為5,上樣濃度為原液濃度,吸附速率為2Bv/h,洗脫速率為1Bv/h。該純化方法簡(jiǎn)單,快捷,純化效率高,進(jìn)一步拓展了刺玫果的應(yīng)用領(lǐng)
