1、生月第7卷第6期ChinJI】gDis(~ectm．icEdition)．Deceni~2314．Ⅷ7．No．6·15··論著·地塞米松通過加強(qiáng)自噬和抑制凋亡減輕海水吸人型肺損傷王莉張敏龍金發(fā)光【摘要】目的探討海水吸入型肺損傷中自噬過程，以及地塞米松對其干預(yù)作用。方法40只大鼠完全隨機(jī)分為正常對照組、1、3、6、12h海水處理組，每組8只。采用氣管內(nèi)滴注海水(3ml／kg)的方法制作海水吸入型急性肺損傷大鼠模型，觀察病理變化，檢測肺組織濕干比，westernblot檢測beclin．1的表達(dá)變化。體外實(shí)驗(yàn)A549細(xì)胞分為正常對照組、12h海水組、地塞米松+1
2、2h海水組、3-MA+12h海水組，采用westernblot檢測beclin一1和bax的表達(dá)變化。結(jié)果海水吸入后，大鼠肺部嚴(yán)重?fù)p傷，水腫明顯，beclin一1隨損傷時間延長表達(dá)量增加，在海水處理后12h達(dá)到高峰。體外實(shí)驗(yàn)中海水刺激增加了beclin．1和bax的表達(dá)，然而與海水組相比，地塞米松預(yù)處理增加了beclin1的表達(dá)，同時使bax的表達(dá)下降。3-MA預(yù)處理則效果相反。結(jié)論自噬參與了海水吸入型肺損傷的修復(fù)過程，地塞米松能夠通過加強(qiáng)自噬并且抑制凋亡減輕肺損傷?！娟P(guān)鍵詞】急性肺損傷；海水；地塞米松；自噬；細(xì)胞凋亡中圖法分類號：R563文獻(xiàn)標(biāo)識碼：ADe
