2、基N(os一cop1)~ORF(開放閱讀框),并克隆到原核表達(dá)載體pET一23d上,將表達(dá)載體pET—Ose—cop1轉(zhuǎn)入大腸桿菌BL21(DE3),以1.0mmol/L的IPTG(isopropyl3-D—thiogalactoside)誘導(dǎo)表達(dá)重組蛋白,然后以重組蛋白作為抗原免疫家兔,制備多克隆抗體。SDS.PAGE電泳分析結(jié)果表明,成功誘導(dǎo)表達(dá)了分子量約為35kD的重組蛋白,Westernblot檢測(cè)表明,免疫家兔的抗血清與水稻幼穗總蛋白雜交信號(hào)較好。Oss—C0P1抗體的制備有助于研究該基因及COPI小泡在水稻中的功能。關(guān)鍵詞Ose—copl水稻,原核表達(dá),抗體P
