2、影響。方法:實(shí)驗(yàn)1:sD雄性大鼠分為對(duì)照組和CCI組。CCI組又分為術(shù)后1d組(Dl組)、術(shù)后3d組(D3組)、術(shù)后7d組(D7組)和術(shù)后14d組(D14組)。分別于術(shù)后1、3、7、14d測(cè)定熱縮足潛伏期(thermalwithdrawallatenc~T、ⅣL)后處死并取其脊髓背角,采用Westernblot方法測(cè)定Akt(P.Akt)蛋白表達(dá)情況。實(shí)驗(yàn)2:sD雄性大鼠分為對(duì)照組、D14’組、D14’+PBS組、D14’+GDNF組。與實(shí)驗(yàn)1相同時(shí)間點(diǎn)測(cè)定TWL,并于第14d處死,取其脊髓背角,采用Westernblot和免疫組化法檢測(cè)P—Akt表達(dá)變化。結(jié)果:
3、與對(duì)照組相比,CCI組大鼠T、vL明顯降低(P<0.01);與D14’組和D14’+PBS組相比,Dl4’+GDNF組大鼠T、ⅣL明顯升高(P<0.01)。CCI組脊髓組織中p-Akt表達(dá)水平于術(shù)后3d開(kāi)始升高,一直持續(xù)到術(shù)后14d(P<0.01):D14’+GDNF組P—Akt表達(dá)水平較D14組和D14’+PBS組顯著下降(P<0.05)。結(jié)論:大鼠脊髓組織中Akt信號(hào)通路可能參與了神經(jīng)病理性疼痛的發(fā)生,鞘內(nèi)注射GDNF減輕神經(jīng)病理性疼痛與降低脊髓組織P—Akt的水平有關(guān)。關(guān)鍵詞膠質(zhì)細(xì)胞源性神經(jīng)營(yíng)養(yǎng)因子;神經(jīng)病理性疼痛;磷酸化AktEFFECTSoFGLlALC
