2、;ELISA法檢測(cè)順鉑分泌IL.8情況;Westernblot法檢測(cè)NF—KB蛋白表達(dá)水平。結(jié)果MTY結(jié)果顯示:與對(duì)照組相比,順鉑濃度為100g/ml對(duì)細(xì)胞增殖有顯著抑制作用(P<0.05)。濃度區(qū)間在200~400g/ml的順鉑對(duì)細(xì)胞的增殖也有抑制作用,該范圍內(nèi)對(duì)細(xì)胞的生長(zhǎng)抑制率無(wú)統(tǒng)計(jì)學(xué)差異(P>0.05)。濃度為100g/ml的順鉑,分別作用于SKOV.3細(xì)胞24h、48h、72h,各時(shí)間組比較無(wú)統(tǒng)計(jì)學(xué)差異(P>0.05)。IL一8(100ng/L)處理細(xì)胞后,SKOV一3細(xì)胞的遷移能力增強(qiáng),順鉑(100g/m1)具有抑制IL一8誘導(dǎo)的
3、SKOV-3細(xì)胞遷移的作用,隨著濃度的增高,遷移的細(xì)胞數(shù)由241.67減少到155.99,差異有統(tǒng)計(jì)學(xué)意義(P<0.05)。順鉑刺激細(xì)胞后,與空白對(duì)照組相比,NF.KB蛋白表達(dá)水平降低(64.04±4.6)。結(jié)論IL-8具有促進(jìn)人卵巢癌SKOV一3細(xì)胞遷移的作用,順鉑可能是通過(guò)NF—KB細(xì)胞信號(hào)通路對(duì)卵巢癌細(xì)胞的遷移起抑制作用?!娟P(guān)鍵詞】卵巢腫瘤;順鉑;白細(xì)胞介素8;細(xì)胞運(yùn)動(dòng)MechanismsofDDPinhibitingcellmigrationinSKOV3cellsinducedbyextraneousIL-8L/Yan—yu,ZH
