1、蠐螬粗提物對(duì)人宮頸癌HeLa細(xì)胞誘導(dǎo)凋亡作用及其機(jī)制的論文【摘要】目的研究蠐螬粗提物對(duì)人宮頸癌hela細(xì)胞誘導(dǎo)凋亡作用，并探討其作用機(jī)制。方法采用流式細(xì)胞儀檢測(cè)hela細(xì)胞的細(xì)胞周期分布相和細(xì)胞凋亡率；末端脫氧核苷酸轉(zhuǎn)移酶（tdt）介導(dǎo)的脫氧核苷酸缺口末端標(biāo)記（tdtmediateddutpnickendlabeling）tunel法檢測(cè)凋亡細(xì)胞；通過(guò)免疫細(xì)胞化學(xué)sp法測(cè)定凋亡相關(guān)基因產(chǎn)物p53、fas、bcl2、bax蛋白的表達(dá)情況。結(jié)果(1)蠐螬粗提物可以誘導(dǎo)hela細(xì)胞發(fā)生凋亡，加藥組出現(xiàn)亞二倍體峰，并且g0/g1期細(xì)胞比例顯著增加（p＜0.01）

2、，而s期和g2/m期細(xì)胞比例則明顯下降,將細(xì)胞阻滯在g0/g1期；（2）蠐螬粗提物作用細(xì)胞后凋亡細(xì)胞數(shù)目隨時(shí)間延長(zhǎng)而增多，凋亡指數(shù)與藥物處理呈時(shí)間依賴性和劑量依賴性；(3)蠐螬粗提物作用后bcl2、p53蛋白隨提取物濃度表達(dá)均下降，bax、fas蛋白表達(dá)上升，四種蛋白表達(dá)各組間比較差異均有具有統(tǒng)計(jì)學(xué)意義（p<0.01）。結(jié)論(1)蠐螬粗提物對(duì)人宮頸癌hela細(xì)胞具有誘導(dǎo)凋亡作用；(2)蠐螬粗提取物誘導(dǎo)凋亡作用機(jī)制可能與細(xì)胞周期發(fā)生g0/g1期阻滯有關(guān)；并且通過(guò)下調(diào)bcl2、p53蛋白表達(dá),上調(diào)bax、fas蛋白表達(dá)，經(jīng)由細(xì)胞凋亡的死亡受體通路和線粒
