2、e中共搜索到1546個(gè)SSR位點(diǎn)袁發(fā)生頻率和平均距離分別是11.99%和15.99kb遙得到的EST-SSR中袁二堿基和三堿基重復(fù)是主要的重復(fù)類型袁分別占30.85%和48.97%袁AT/TA和GAA/CTT分別是二堿基和三堿基的優(yōu)勢(shì)重復(fù)類型遙對(duì)長(zhǎng)度逸20bp的SSR共設(shè)計(jì)合成了56對(duì)引物袁在這10個(gè)材料中進(jìn)行檢測(cè)袁能擴(kuò)增出清晰條帶的有38對(duì)袁占67.86%袁其中呈多態(tài)性的有9對(duì)袁占23.68%袁并對(duì)其所在的Unigene功能和表達(dá)量進(jìn)行了初步分析遙本研究進(jìn)一步證明了棉花EST-SSR標(biāo)記開(kāi)發(fā)的可行性袁并且為棉花高密度遺傳圖譜和腺體相關(guān)基因的研究奠定了基礎(chǔ)遙關(guān)鍵詞院棉花曰EST-SSR標(biāo)記曰多

5、rmationfrom10glandandglandlessmaterials.1546EST-SSRswereobtainedfrom12895unigenesmorethan1kbinlength,whichaccountsfor11.99%,andtheaveragedistanceofSSRswas15.99kb.AmongtheobtainedEST-SSRs,dinu-cleotideandtrinucleotiderepeatswerethedominanttypes,accountingfor30.85%and48.97%respectively.AT/TAandGAA/CTT
6、werethemostabundantmotifsrespectivelyindinucleotideandtrinucleotiderepeats.Totalof56pairsofprimersweredesignedandsynthesizedfortheSSRsmorethan20bpandtestedinthe10materialstoevaluatetheapplicationandpoly-morphism.38pairsofprimerscouldproduceclearbandsinPAGEelectrophoresis,accountingfor67.86%,and9EST-
7、SSRsa-mongthemshowedpolymorphism,accountingfor23.68%.Furthermore,thefunctionsandexpressionoftheunigenescontain-ingpolymorphicEST-SSRwereanalyzedpreliminarily.Thisstudyfurtherprovesthatitisfeasibletode
