1、大鼠胎血間充質(zhì)干細(xì)胞移植對(duì)局灶性腦缺血治療作用的研究【摘要】目的:研究大鼠胎血間充質(zhì)干細(xì)胞(MSCs)移植入大腦中動(dòng)脈栓塞(MCAO)大鼠模型后大鼠神經(jīng)功能的恢復(fù)情況。方法:線栓法建立左側(cè)大鼠MCAO模型,隨機(jī)分為3組:手術(shù)組、MSCs移植組、生理鹽水組。5溴2脫氧尿核苷(BrdU)標(biāo)記的MSCs移植后,采用改良神經(jīng)功能損傷評(píng)分系統(tǒng)(mNSS)評(píng)價(jià)大鼠神經(jīng)功能恢復(fù)情況;應(yīng)用免疫組織化學(xué)雙染技術(shù)檢測(cè)MSCs的存活、遷移及其巢蛋白(Nestin)、膠質(zhì)纖維酸性蛋白(GFAP)和神經(jīng)元特異性烯醇化酶(N
2、SE)的表達(dá)。結(jié)果:MSCs增殖能力強(qiáng),并能在體外誘導(dǎo)分化為成骨細(xì)胞、脂肪細(xì)胞和神經(jīng)元樣細(xì)胞。MSCs移植后顯著提高局灶性腦缺血大鼠神經(jīng)功能的恢復(fù)。移植的MSCs細(xì)胞可在大鼠腦組織中存活,并向缺血區(qū)域遷移,有不同比例MSCs細(xì)胞分別表達(dá)Nestin、GFAP、NSE。結(jié)論:大鼠胎血中含有MSCs,體外擴(kuò)增純化后可促進(jìn)局灶性腦缺血大鼠神經(jīng)功能的恢復(fù),移植細(xì)胞可在大鼠腦缺血區(qū)域中存活、遷移并向星形膠質(zhì)細(xì)胞或神經(jīng)元分化?!娟P(guān)鍵詞】胎血干細(xì)胞缺氧缺血腦移植大鼠WistarATransplantationof
