1、摘要本文從珠江三角洲地區(qū)感染豬群進(jìn)行豬傳染性胸膜肺炎放線桿菌的分離鑒定及血清學(xué)定型,己分離到7株APP野毒株,經(jīng)培養(yǎng)特性觀察、“衛(wèi)星”現(xiàn)象、CAMP試驗(yàn)及糖發(fā)酵、尿索酶試驗(yàn)均符合豬傳染性胸膜肺炎放線桿菌生物學(xué)特性,在PCR鑒定中均可擴(kuò)增出APPapxIVA基因大小約為442bp的特異核苷酸片段,動(dòng)物試驗(yàn)表明7株APP分離菌株均具致病性,血清分型試驗(yàn)表明有3株為血清I型APP,2株為血清7型APP,血清3、4型各為l株。對(duì)珠江三角洲地區(qū)31個(gè)集約化豬場(chǎng)共450份血清進(jìn)行IHA試驗(yàn)檢測(cè),結(jié)果顯示APP血清抗體陽性率為58.4%,經(jīng)血清分型鑒定表明該地區(qū)集約化豬場(chǎng)主
2、要流行的血清型為APP1型,其次為APP7型和APP3型。以2株強(qiáng)毒株為地方適用型滅活疫苗菌種,探索了滅活疫苗生產(chǎn)的最佳營養(yǎng)要索及培養(yǎng)環(huán)境,在各種最佳優(yōu)化條件下培養(yǎng)出的APP菌液濃度可達(dá)菌數(shù)24億/m1,經(jīng)安全性試驗(yàn)、理化性狀檢測(cè)表明疫苗安全有效。對(duì)兔及豬進(jìn)行免疫,其抗體消長規(guī)律顯示油乳佐劑滅活苗比鋁膠佐劑滅活苗的免疫源性好,適時(shí)進(jìn)行二次免疫,會(huì)顯著提高抗體的水平和有更長的保護(hù)期。攻毒試驗(yàn)表明二價(jià)滅活苗對(duì)兩個(gè)型的APP都具有可靠保護(hù)。疫苗田聞試驗(yàn)表明免疫后豬群料肉比低、上市重高、上市日齡短,死亡率及殘次率較低。顯示制備的APP雙價(jià)地方適用型滅活疫苗可在臨床推廣
