2、漸增大、形態(tài)往上皮樣轉(zhuǎn)變,胞漿豐富,出現(xiàn)雙核或多核細胞。RT—PCR和免疫熒光檢測結(jié)果顯示,AFP在誘導初期表達,在誘導后期表達下降;CKl8、ALB和TAT的表達與誘導時間呈線性關系。第20天達到表達高峰;誘導20d后免疫熒光檢測,AFP、CKl8、ALB和TAT均為陽性;誘導20d細胞的PAS糖原合成反應呈陽性,即具備糖原合成和儲存的肝細胞特有功能。結(jié)論HGF、FGF-4和OSM三種細胞因子組合,可誘導小鼠問充質(zhì)干細胞向肝細胞分化,該結(jié)果為間充質(zhì)干細胞跨越分化肝細胞的分子機制探討和臨床應用打下了基礎?!娟P鍵詞】間充質(zhì)干細胞
