1、第28卷第11期生態(tài)學(xué)報(bào)Vol.28,No.112008年11月ACTAECOLOGICASINICANov.,2008基于ISSR標(biāo)記的扁玉螺(Neveritadidyma)自然居群遺傳結(jié)構(gòu)3孫始威,孫振興,葛宜和,閆冬春,黃清榮(魯東大學(xué)生命科學(xué)學(xué)院,煙臺(tái)264025)摘要:利用ISSR分子標(biāo)記技術(shù),對(duì)采自大連(DL)、煙臺(tái)(YT)及青島(QD)近海的扁玉螺3個(gè)自然居群的遺傳結(jié)構(gòu)和遺傳多樣性進(jìn)行了分析。用13個(gè)引物對(duì)90只個(gè)體進(jìn)行了PCR擴(kuò)增,共檢測(cè)到161個(gè)位點(diǎn),3個(gè)居群的多態(tài)位點(diǎn)比例為74.53%~
2、85109%,各居群遺傳多樣性水平的高低依次為YT>QD>DL。扁玉螺在物種水平上的Nei’s基因多樣性指數(shù)和Shannon’s信息指數(shù)分別為0.3395和0.5113,在居群水平上分別為0.2811和0.4189,顯示出扁玉螺有著較高的遺傳多樣性。AMOVA分子變異分析表明,扁玉螺的遺傳變異有27.16%發(fā)生在居群間,72.84%發(fā)生在居群內(nèi),居群內(nèi)的遺傳變異大于居群間的遺傳變異。扁玉螺3個(gè)居群間的遺傳分化系數(shù)(Gst)為0.1720,基因流(Nm)為2.4063,Nei’s遺傳距離平均值為0.1228,
