2、磊(1.江蘇省農(nóng)業(yè)科學(xué)院蔬菜研究所,江蘇南京210014;2.江蘇省農(nóng)業(yè)科學(xué)院農(nóng)業(yè)生物技術(shù)研究所,江蘇南京210014)搖搖摘要:搖本研究將已構(gòu)建的SUC2鄄GroEL和pBI121鄄AC1鄄AC2植物表達(dá)載體共轉(zhuǎn)化番茄,共獲得19株卡那霉素抗性再生植株。經(jīng)過(guò)PCR和RT鄄PCR檢測(cè),初步獲得轉(zhuǎn)SUC2鄄GroEL載體的陽(yáng)性植株5株,轉(zhuǎn)pBI121鄄AC1鄄AC2載體的陽(yáng)性植株3株以及轉(zhuǎn)SUC2鄄GroEL/pBI121鄄AC1鄄AC2雙載體的陽(yáng)性植株2株。利用TYLCV侵染性克隆農(nóng)桿菌注射接種法對(duì)T代轉(zhuǎn)基
3、因陽(yáng)性植株進(jìn)行抗性鑒定,獲得抗TYLCV的轉(zhuǎn)基因植株7株。對(duì)T代未擴(kuò)出病毒條帶的T001代植株進(jìn)行抗性鑒定,結(jié)果發(fā)現(xiàn)轉(zhuǎn)SUC2鄄GroEL、pBI121鄄AC1鄄AC2、SUC2鄄GroEL/pBI121鄄AC1鄄AC2不同載體的株系植株中平均帶毒率依次是35%、25%和15%,推斷轉(zhuǎn)雙基因的植株的抗病能力優(yōu)于轉(zhuǎn)單個(gè)基因植株的抗病能力。關(guān)鍵詞:搖番茄黃化曲葉病毒;RNAi;GroEL;共轉(zhuǎn)化;轉(zhuǎn)基因番茄+中圖分類號(hào):搖S436.412.17搖搖搖文獻(xiàn)標(biāo)識(shí)碼:搖A搖搖搖文章編號(hào):搖1000鄄4440(2014
