2、肌細胞缺氧/復氧損傷的保護作用。方法采用高效液相色譜法,色譜柱:KromasilC18-1柱(4.6mm伊250mm,5滋m);流動相:以甲醇-水(70頤30)洗脫,流速為1.0mL·min,檢測波長為280nm,柱溫室溫,進樣量:10滋L。并用原代培養(yǎng)的乳鼠心肌細胞建立缺氧復/氧損傷模型,異芹菜腦在建模前進行預干預,心肌細胞隨機分為5組,空白對照組(Control,CON),缺氧/復氧組(hypoxia/reoxygenation,H/R),異芹菜腦低、中、高劑量干預組(IsoL、IsoM、IsoH)。缺氧4h/復氧4h后,采用CCK-8法檢測心肌細胞
3、存活-1率。結果異芹菜腦在0.02096~0.2096mg·mL線性關系良好(r=0.9999),平均回收率為98.09%,RSD=1.25%。與空白對照組比較,缺氧/復氧組心肌細胞存活率顯著降低(P約0.01);與缺氧/復氧組比較,IsoL、IsoM、IsoH劑量干預組心肌細胞存活率均顯著升高。結論異芹菜腦對體外培養(yǎng)乳鼠心肌細胞缺氧/復氧損傷具有一定的保護作用,隔山香藥材中異芹菜腦的HPLC測定方法準確、簡便、重現(xiàn)性好,有助于完善隔山香藥材的質量控制。關鍵詞:異芹菜腦;含量測定;高效液相色譜法;心肌細胞;缺氧/復氧損傷中圖分類號:R284.1;285.
