1、中華實驗外科雜志2014年9月第31卷第9期ChinJExpSurg，September2014，Vo1．31．No．9·l933··實驗研究·體內(nèi)微環(huán)境對肝卵圓細(xì)胞分化的影響李常海廖鉑王艷軍董為董漢華梁慧芳陳孝平【摘要】目的研究體內(nèi)微環(huán)境對肝卵圓細(xì)胞分化的影響，探討肝卵圓細(xì)胞是否可在體內(nèi)分化為肝癌。方法在使用黃曲霉素作為誘癌劑的情況下，將轉(zhuǎn)染乙肝病毒x基因(HBx)的大鼠卵圓細(xì)胞種植于裸鼠皮下，形成腫瘤后再移植至同一裸鼠肝實質(zhì)內(nèi)，使其繼續(xù)生長。收集皮下腫瘤和肝內(nèi)腫瘤，用蘇木素一伊紅(HE)、免疫組織化學(xué)染色和透射電鏡對其進(jìn)行檢查，明確其性質(zhì)。結(jié)果10周時，實驗組中10只(1
2、0／15)裸鼠形成皮下腫瘤，對照組中8只(8／15)裸鼠形成皮下腫瘤，兩組在腫瘤大小和生長速度方面差異無統(tǒng)計學(xué)意義(P>0。05)；兩組均在皮下分化成間質(zhì)腫瘤，表達(dá)間質(zhì)性標(biāo)志物波形蛋白(Vimentin)和平滑肌肌動蛋白(SMA)。皮下問質(zhì)腫瘤移植至肝內(nèi)8周后，在肝內(nèi)種植處長成肝內(nèi)腫瘤；經(jīng)病理確診實驗組腫瘤為肝癌肉瘤(由惡性間質(zhì)細(xì)胞和肝癌細(xì)胞組成)，表達(dá)HepParl、細(xì)胞角蛋白8(CK8)和甲胎蛋白(AFP)抗原，說明肝卵圓細(xì)胞在HBx和黃曲霉素的共同作用下可部分分化成肝細(xì)胞癌。結(jié)論肝卵圓細(xì)胞具有可塑性，局部微環(huán)境對其分化方向起關(guān)鍵作用；肝癌可能來源于卵圓細(xì)胞的異常分化?！?/p>
