1、nCT●ilglGUUUHAEBD咖LI-SIHIGli華北農學報·2005,20(5):1.9通過基因槍和農桿菌介導用BADH基因轉化小麥RAZZAQAbdull’2,張艷敏1,趙和1,馬峙英2,郭北海1,王海波1(1.河北省農林科學院遺傳生理研究所,河北石家莊050051;2.河北農業(yè)大學,河北保定071001)摘要:土壤鹽堿是一種嚴重障礙作物生產的環(huán)境因子,甜菜堿醛脫氫酶BADH基因是一種重要的可賦予植物滲透調節(jié)抗性的基因。本研究用基因槍法及農桿菌介導法向小麥幼胚和成熟胚愈傷組織導入了BADH基因。用PDS一1000/HE基因槍轟擊2933塊幼胚愈傷組織,
2、分化出了45株再生植株,分化率為1.53%。PCR分析表明,其中的5株為BADH基因轉化植株。用PFY涂抹其葉片,進一步證實了PCR的結果。以小麥成熟胚愈傷組織為受體,用農桿菌介導轉化1968塊愈傷,僅再生出了5株綠苗,PCR檢測結果均為陰性。但對其轉化愈傷組織的PCR檢測表明,外源基因已在受體細胞中實現(xiàn)了整合。以幼胚愈傷組織為受體,用農桿菌介導轉化2933塊愈傷,共再生出了21株綠苗。對其進行PCR檢測,僅有5株為BADH基因轉化植株。轉化處理過的幼胚愈傷組織的綠苗再生率(0.72%)高于成熟胚愈傷(0.25%)。與對照相比,所有的轉化植株均能夠在0.5%Na
