1、nCT●ilglGUUUHAEBD咖LI-SIHIGli華北農(nóng)學(xué)報(bào)·2005,20(5):1.9通過基因槍和農(nóng)桿菌介導(dǎo)用BADH基因轉(zhuǎn)化小麥RAZZAQAbdull’2,張艷敏1,趙和1,馬峙英2,郭北海1,王海波1(1.河北省農(nóng)林科學(xué)院遺傳生理研究所,河北石家莊050051;2.河北農(nóng)業(yè)大學(xué),河北保定071001)摘要:土壤鹽堿是一種嚴(yán)重障礙作物生產(chǎn)的環(huán)境因子,甜菜堿醛脫氫酶BADH基因是一種重要的可賦予植物滲透調(diào)節(jié)抗性的基因。本研究用基因槍法及農(nóng)桿菌介導(dǎo)法向小麥幼胚和成熟胚愈傷組織導(dǎo)入了BADH基因。用PDS一1000/HE基因槍轟擊2933塊幼胚愈傷組織,
2、分化出了45株再生植株,分化率為1.53%。PCR分析表明,其中的5株為BADH基因轉(zhuǎn)化植株。用PFY涂抹其葉片,進(jìn)一步證實(shí)了PCR的結(jié)果。以小麥成熟胚愈傷組織為受體,用農(nóng)桿菌介導(dǎo)轉(zhuǎn)化1968塊愈傷,僅再生出了5株綠苗,PCR檢測結(jié)果均為陰性。但對其轉(zhuǎn)化愈傷組織的PCR檢測表明,外源基因已在受體細(xì)胞中實(shí)現(xiàn)了整合。以幼胚愈傷組織為受體,用農(nóng)桿菌介導(dǎo)轉(zhuǎn)化2933塊愈傷,共再生出了21株綠苗。對其進(jìn)行PCR檢測,僅有5株為BADH基因轉(zhuǎn)化植株。轉(zhuǎn)化處理過的幼胚愈傷組織的綠苗再生率(0.72%)高于成熟胚愈傷(0.25%)。與對照相比,所有的轉(zhuǎn)化植株均能夠在0.5%Na
