1、清開靈干預(yù)新生大鼠缺氧缺血腦皮質(zhì)白介素-6及Fas-L蛋白的實(shí)驗(yàn)研究作者:陳敬國,方慧云,蔣犁,程敏婷【關(guān)鍵詞】腦缺氧;,,腦缺血;,,新生大鼠;,,清開靈;,,白介素-6;,,Fas-L蛋白 摘要:目的探討缺氧缺血腦損傷對新生大鼠HIBD腦皮質(zhì)IL-6放射免疫濃度,F(xiàn)as-L蛋白表達(dá)的影響,及清開靈對新生大鼠HIBD的干預(yù)治療作用以及有關(guān)分子機(jī)制。方法建立HIBD動物模型后分為放射免疫法測定組和免疫組化測定組,每組又分為清開靈干預(yù)組、生理鹽水對照組、假手術(shù)組,根據(jù)檢測需要,在缺氧缺血后24h,3d,7d作IL-6腦皮質(zhì)勻漿放射免疫濃度測定、Fas-L蛋白免疫組
2、化SP法測定。結(jié)果①腦皮質(zhì)組織勻漿IL-6放射免疫濃度(pg/ml)假手術(shù)組、生理鹽水組、清開靈干預(yù)組:24h依次為50.1±6.2,56.3±7.1,51.3±6.8;3d依次為50.6±6.4,250.3±11.2,78.1±8.3;7d依次為51.4±6.6,345.7±12.3,81.2±9.2,生理鹽水組與另兩組分別比較,24hP均>0.05,無明顯變化;3,7d生理鹽水組增加明顯P<0.05)清開靈干預(yù)組降低明顯P<0.05)。②③④左腦皮質(zhì)Fas-L蛋白表達(dá)陽性率假手術(shù)組、生理鹽水組、清開靈干預(yù)組:24h依次為8.04±3.50,120.
3、20±6.10,21.10±5.10;3d依次為8.20±3.10,87.6±5.90,13.40±4.20;7d依次為8.01±2.80,8.32±112.90,8.26±2.83;生理鹽水組與另兩組分別比較,7dP均>0.05,無明顯變化;24h,3d增多明顯(P<0.05),清開靈干預(yù)組降低明顯(P<0.05)。結(jié)論①新生大鼠HIBD后24h,3d,7d腦皮質(zhì)IL-6含量逐漸增高。②清開靈能降低新生大鼠HIBD腦皮質(zhì)IL-6的產(chǎn)生量。③新生大鼠HIBD后,結(jié)扎側(cè)腦皮質(zhì)fas-L蛋白表達(dá)增高。④清開靈能抑制新生大鼠HIBD結(jié)扎側(cè)腦皮質(zhì)fas-L蛋白
