2、總RNA,采用SMART和LD.PcR合成全長(zhǎng)的雙鏈cDNA(dscDNA),并對(duì)其進(jìn)行均一化和SfiI酶切處理。處理后的dscDNA分另II連接3種閱讀框pGADT7.Sfi威體,將連接產(chǎn)物轉(zhuǎn)化至大腸桿菌BH5a構(gòu)建三框eDNA初級(jí)文庫(kù),取三框eDNA初級(jí)文庫(kù)進(jìn)行混合擴(kuò)增,通過(guò)滴度測(cè)定和PCR鑒定其庫(kù)容量和基因重組率,最后收集文庫(kù)細(xì)菌提取文庫(kù)質(zhì)粒?!窘Y(jié)果】3種讀碼框eDNA文庫(kù)的庫(kù)容量分別為3.0×106.2.OxlOe和2.0×106CFU,文庫(kù)實(shí)際擴(kuò)增基數(shù)>150萬(wàn)CFU;一號(hào)和三號(hào)讀碼框的基因重組率均為100%,二號(hào)讀碼框的基因重組率大于93%。eDN
