6、TA10ml,5MNaCl20ml,加滅菌蒸餾水1000ml。終濃度:10mMTris,100mMNaCl,5mMEDTA]緩沖液3ml、SDS(十二烷基硫酸鈉,10%)溶液0.3ml和選用的酶溶液200μl,置45℃恒溫箱內(nèi)觀察3~6h,待液體近透明(無明顯固體)時(shí),加入3ml蒸餾水,混勻后離心、去上清涂片鏡檢。1.3.3強(qiáng)酸消化法取B組、C組和D組大鼠的肺臟、肝臟、腎臟及脾臟等臟器,按文獻(xiàn)報(bào)道所示步驟進(jìn)行消化[2]。1.3.4酶選擇實(shí)驗(yàn)筆者應(yīng)用了市面上常見且價(jià)格合理的8種生物酶進(jìn)行了選擇性試驗(yàn),取A
7、組大鼠的各種臟器,剪碎后分別加入有標(biāo)記的試管中,每只試管加入一種酶(濃度暫定為20mg/ml),按1.3.2所示步驟進(jìn)行消化。1.3.5消化靈敏度實(shí)驗(yàn)按1.3.4步驟將選擇后酶的濃度分成40mg/ml、20mg/ml、10mg/ml和5mg/ml,取A組大鼠的肝臟、肺臟、脾臟及腎臟,按1.3.2所示步驟進(jìn)行消化。1.3.6酶組合消化臟器試驗(yàn)取B組、C組和D組大鼠的肝臟、肺臟、脾臟及腎臟等,根據(jù)1.3.4和1.3.5的結(jié)果選擇幾種酶類配制一定濃度后按1.3.2所示步驟進(jìn)行消化。2結(jié)果通過肉眼觀察,將各種酶
8、的消化能力分成4級,分別用羅馬數(shù)字Ⅰ~Ⅳ級表示。Ⅰ級表示消化能力最強(qiáng),Ⅳ級表示消化能力最弱,具體意義如下:Ⅰ級:液體中可見少許固體物質(zhì),約10%;Ⅱ級:液體中剩約25%的固體物質(zhì);Ⅲ級:液體中剩約50%的固體物質(zhì);Ⅳ級:液體中剩約75%的固體物質(zhì)。2.1酶選擇試驗(yàn)見表1。從表1中可以看出:(1)肺:胰蛋白酶、蛋白酶K和膠原酶的消化能力均達(dá)到Ⅰ級,膠原酶/分散酶和透明質(zhì)酸酶消化肺臟的消化能力可達(dá)到Ⅱ級;(2)肝:蛋白酶K和膠原酶Ⅱ的消化能力均
