2、GEM-T載體,構(gòu)建重組質(zhì)粒pGEM-T-poIFN-α1,進行測序。以pGEM-T-poIFN-α1為模板,經(jīng)PCR擴增帶有酶切位點的poIFN-α1基因,將其定向克隆到原核表達載體pGEX-6P-1中,構(gòu)建重組表達載體pGEX-6P-poIFNα1,轉(zhuǎn)化大腸桿菌BL21(DE3)進行蛋白誘導表達和鑒定,并確定蛋白最佳表達條件。序列分析表明,克隆的poIFN-α1基因全長546bp,編碼181個氨基酸,前23個氨基酸組成信號肽,無糖基化位點。SDS-PAGE和Westernblotting證實重組表達菌經(jīng)IPTG誘導
