1、天津大學(xué)碩士學(xué)位論文11β-羥基孕甾-1,4,16-三烯-3,20-二酮合成過程研究姓名:苑文博申請學(xué)位級別:碩士專業(yè):化學(xué)工程指導(dǎo)教師:王永莉20070601中文摘要地塞米松屬于孕甾烷類腎上腺皮質(zhì)激素,具有影響糖代謝、抗炎、抗過敏、抗毒素等作用。1lB.羥基孕甾.1,4,16.三烯.3,20.二酮(HTD)是生產(chǎn)地塞米松的一種中間體,直接影響著地塞米松質(zhì)量和成本。為提高該物質(zhì)合成過程的轉(zhuǎn)化率與收率,本文對HTD的反應(yīng)合成過程進(jìn)行了較系統(tǒng)地研究。對HTD的反應(yīng)合成進(jìn)行了研究,考察了反應(yīng)溫度、金屬鉻顆粒的大小、鹽酸濃度、物料配比、氮?dú)饬魉?、攪拌?qiáng)度、溶劑A使用量和時(shí)間對反應(yīng)過
2、程轉(zhuǎn)化率和收率的影響,初步確定了經(jīng)驗(yàn)最佳的合成工藝。采用分光光度法對鉻與稀鹽酸的流固相非催化反應(yīng)進(jìn)行了測定,探討反應(yīng)的控制步驟,并得到了相應(yīng)的反應(yīng)動力學(xué)方程,從而確定出影響反應(yīng)速度的關(guān)鍵因素。HTD是以16a.,17a.氧環(huán).1l咖羥基孕甾.1,4啊二烯.3,20.二酮(以下簡稱EHDD)為原料合成的,由于HTD與EHDD分子與晶體結(jié)構(gòu)比較相似,本文還提出了HTD和EHDD在溶液結(jié)晶過程中可能形成固體溶液型混晶的假設(shè)。通過設(shè)計(jì)實(shí)驗(yàn)制備出了不同質(zhì)量比的HTD/EHDD混晶產(chǎn)品,并采用XRD、DSC以及HPLC等表征手段證明了該假設(shè)的正確性。上述研究結(jié)果可為HTD合成過程的進(jìn)一
