1、河北醫(yī)藥2017年4月第39卷第8期HebeiMedicalJournal,2017,Vol39AprNo.81141doi:10.3969/j.issn.1002-7386.2017.08.005·論著·龍膽水提物對急性胰腺炎大鼠肝損傷的保護(hù)作用及機制研究吳軍尚瑞【摘要】目的探討龍膽水提物對急性胰腺炎(AP)大鼠肝損傷的保護(hù)作用及機制。方法SD大鼠40只隨機分為正常組(n=8)與模型組(n=32),模型組采用經(jīng)十二指腸乳頭逆行膽胰管注射5%牛黃膽酸鈉方法制備AP大鼠模型,造模后大鼠分為4個亞組:模型對照組(n=8)、龍膽水提物12mg/kg
2、組(n=8)、24mg/kg組(n=8)、48mg/kg組(n=8),龍膽水提物低、中、高劑量組每天分別灌胃給予龍膽水提物12mg/kg,24mg/kg及48mg/kg,正常組和模型對照組給予等體積的0.9%氯化鈉溶液,連續(xù)給藥1周,1次/d。給藥結(jié)束后頸總動脈取血,分離血清,檢測血清ALT、AST、AMY、IL-6、TNF-α水平,大鼠脫頸椎處死后剪取肝右葉1/2,于4%中性甲醛固定,HE染色觀察肝臟結(jié)構(gòu)變化,westernblot檢測肝臟組織NF-κβ及I-κB表達(dá)水平。結(jié)果與模型對照組比較,龍膽水提物24mg/kg及48mg/kg組血清
