2、:成年雄性SD大鼠48只,隨機(jī)分為4組:正常組(不做任何處理);SCS組(植入SCS裝置并給予SCS刺激);SNL+shamSCS組(給予SNL手術(shù)并植入SCS裝置,但不進(jìn)行刺激);SNL+SCS組(SNL手術(shù)并給予SCS刺激)。SCS刺激是在SNL術(shù)后第6—10d進(jìn)行(8h/d),第10d刺激結(jié)束后處死動(dòng)物。運(yùn)用行為學(xué)方法檢測(cè)慢性痛狀態(tài)下大鼠后肢對(duì)機(jī)械性刺激的反應(yīng)閾值;采用免疫組織化學(xué)染色和Westernblot方法分別檢測(cè)脊髓背角內(nèi)NR2B和星形膠質(zhì)細(xì)胞的標(biāo)志物GFAP的表達(dá)變化。結(jié)果:(1)SNL術(shù)后大鼠手術(shù)側(cè)后足機(jī)械性痛敏顯著增加,第6~10d給予SCS刺激后,可觀察到大鼠的
3、痛行為學(xué)表現(xiàn)有明顯緩解;(2)免疫組化結(jié)果顯示:與SNL+shamSCS組相比,SNL+SCS組大鼠脊髓背角內(nèi)NR2B和GFAP免疫陽(yáng)性細(xì)胞的數(shù)量顯著減少;(3)Westernblot結(jié)果顯示:給予SCS刺激后,SNL大鼠腰膨大段脊髓背角內(nèi)NR2B的表達(dá)量顯著下調(diào),同時(shí)GFAP的表達(dá)量也明顯有所降低。結(jié)論:給予SCS刺激可以有效地緩解SNL模型大鼠的神經(jīng)病理性痛的行為學(xué)表現(xiàn);該作用可能與SCS刺激抑制脊髓背角內(nèi)NR2B的表達(dá)和星形膠質(zhì)細(xì)胞的激活密切相關(guān)?!娟P(guān)鍵詞】脊髓刺激術(shù);神經(jīng)病理性痛;NR2B;星形膠質(zhì)細(xì)胞;大鼠TheeffectofSCSonNR2Breceptorandthe
