2、,用間接ELISA方法和免疫印跡進(jìn)行篩選,然后對(duì)篩選出來的細(xì)胞株進(jìn)行一般性質(zhì)測定、親和常數(shù)測定、Ic測定。結(jié)果:合成多肽純度>95%,細(xì)胞融合后,其平均融合率為75%,并用間接ELISA方法和Westernblot篩選出1條可與FOXL2蛋白發(fā)生明顯反應(yīng)的細(xì)胞株c2;經(jīng)測定,c2細(xì)胞株上清效價(jià)為1/128,腹水效價(jià)為1/256,其亞型屬于IgGl,雜交瘤細(xì)胞染色體為102條,腹水單抗的蛋白含量為1.576g/L,其親和常數(shù)為2.12×10L/mol,IC值為0.08247L。結(jié)論:成功制備了FOXL2蛋白單克隆抗體,為下一步深入研究FOXL2蛋白奠定基
