1、人小細(xì)胞肺癌細(xì)胞H446Snail基因表達(dá)及其在順鉑耐藥中的作用況里杉,周向東(400038重慶,第三軍醫(yī)大學(xué)西南醫(yī)院呼吸內(nèi)科)[摘要] 目的觀察鋅指轉(zhuǎn)錄因子snail在耐順鉑(cisplatin,CDDP)的人小細(xì)胞肺癌(smallcelllungcancer,SCLC)H446/CDDP細(xì)胞中的表達(dá)情況,探討snail表達(dá)在人SCLC順鉑耐藥中的作用。方法 培養(yǎng)H446細(xì)胞系和H446/CDDP細(xì)胞系,采用MTT法檢測H446/CDDP的耐藥指數(shù);分別提取人SCLC順鉑耐藥細(xì)胞H446/CDDP及其親代細(xì)胞H446的mRNA及總蛋白,應(yīng)用sybgree

7、間充質(zhì)轉(zhuǎn)化(epithelialtomesenchymaltransition,EMT)的發(fā)生,進而賦予上皮源性腫瘤細(xì)胞運動和侵襲能力[2,3]。在肺腺癌細(xì)胞系A(chǔ)549的研究中發(fā)現(xiàn)沉默Snail的表達(dá)能夠恢復(fù)細(xì)胞對順鉑的敏感性[4,5],而在SCLC尚缺乏相關(guān)方面的研究。因此,本研究擬從EMT的關(guān)鍵因子snail入手,研究人SCLC順鉑耐藥性的獲得與snail表達(dá)水平之間的關(guān)系,為逆轉(zhuǎn)其耐藥尋求新的途徑。[基金項目] 國家重點基礎(chǔ)研究發(fā)展計劃(973計劃,2010CB529402)[通信作者] 周向東,E-mai:xiangdongzhou@126.com
