1、廣西農(nóng)業(yè)科學(xué)2010,41(6):598-601GuallgxiAgriculturalSciences豬D型產(chǎn)毒素多殺性巴氏桿菌毒素分段基因的原核表達(dá)黃百花,劉文娟,李志勇,譚春萍,周松峰,譚穎翌,羅佳,陸芹章+(廣西大學(xué)動物科學(xué)技術(shù)學(xué)院,南寧530005)摘要:根撤,enBarrk已發(fā)表的產(chǎn)毒素多殺性巴氏桿菌(T.Plll)f0】A基因序列(A腳78)設(shè)計(jì)1對引物,從豬源D型'PPm中擴(kuò)增出toxA基因片段(3858bp)。再根據(jù)toxA基因序列設(shè)計(jì)3對引物,分別擴(kuò)增出ZQ(1084bp)、ZH(1092bp)、HM(9
2、06bp)3個分段基因,將其克隆至原核表達(dá)載體pET32a,轉(zhuǎn)入大腸桿菌BL21中進(jìn)行融合表達(dá)。經(jīng)SDS—PAGE和West—emblotting檢測分析,發(fā)r-覓ZQ、HM基因的表達(dá)產(chǎn)物以包涵體形式存在,大小分別為75、50kDa,ZH基因不表達(dá)。動物試驗(yàn)結(jié)果表明,HM重組蛋白具有良好的免疫原性、反應(yīng)原性及一定的生物學(xué)活性,而ZQ重組蛋白沒有免疫原性、反應(yīng)原性及生物學(xué)活性。通過間接ELlSA的方法檢測抗毒素的陽性豬血清,結(jié)果表明,HM重組蛋白較天然蛋白具有更高的特異性。關(guān)鍵詞:豬D型產(chǎn)毒素多殺巴氏桿菌;毒素基因;分段;原核
