1、醫(yī)護職稱的名與實評定職稱,有一個名與實的問題。關于名實的涵袁媛,司維柯,李招權,潘靜,趙宸【摘要】本研究探討外源性Wnt5a誘導K562細胞定向分化的作用。用重組腺病毒AdWnt5a、AdGFP感染CHO細胞,分別制備Wnt5a和GFP對照條件培養(yǎng)液,并處理K562細胞H7天,用光學顯微鏡和電子顯微鏡觀察細胞形態(tài)變化;采用過氧化物酶、糖原和非特異性酯酶染色、細胞表面分化抗原CD13、CD14、CD68鑒定K562細胞的分化表型;用流式細胞術檢測細胞分化周期的變化。結果表明:與對照GFP條件培養(yǎng)液相比,Wnt5a條件培養(yǎng)液處理的K562細胞形態(tài)和超微結構均出現(xiàn)了分化成熟的特征;POX、PAS
2、、aNAE細胞化學染色均為陽性,并且aNAE陽性70°/??杀环c抑制;單核細胞系表面標志抗原CD14、CD68和粒細胞系表面標志抗原CD13經(jīng)免疫細胞化學染色均呈陽性,但CD13與對照比較無顯著性差異;Wnt5a處理K562細胞5天,細胞周期阻滯在G2期。結論:外源性的Wnt5a可誘導K562細胞向成熟方向分化,并且有向單核細胞系分化的現(xiàn)象?!娟P鍵詞】誘導分化DifferentiationPhenotypesofK562CellsInducedbyExogenousWnt5aAbstractThisstudywasaimedtoinvestigatetheeffectofexogeno

5、aNAEandimmtheeffectofWnt5aotionofK562cellswaometry.TheresultsslogyandultrastructatedbyWnt5adisplayaturefeature;bothhowedhigherposititedgroupthanthatincontrolgroup;theaNAEstainingalsowaspositive,butpositiveintensityinWnt5atreatedgroupcouldbeinhibitedupto70%byNaF.Theexpressionsofmonocyticdifferentia
6、tionantigensofCD14,CD68inWnt5atreatedgroupwerehigherthanthoseincontrolgroup,buttheexpressiondifferencesofgranulocytiedifferentiationantigenCDl3betweenWnt5atreatedgroupandcontrolgroupwerenotsignificant.ThecellcycleintreatedgroupwasblockedatG2phaseascomparedwithcontrolgroup.Itisconcludedthatexogenou
