4、oryratewas43.0%,54.9%,78.5%respectively.Thecharacteristicmorphologicalchangesofapoptosiswereobservedincellsaftertreatedby2mg/LofTaoerqi,andmanymultinucleategiantcellswerealsofound?TaoerqiinducedaG2/Mcellcyclearrestandtheapoptosiswastimeanddosedependent.Withthetreat
5、mentof2mg/LTaoerqifor48h,theproteinexpressionsofPCNAandBcl2decreasedfrom(154.78±0.16)and(85.56±0.09)to(7&46±0」5)and(40.43±0.07)(P0.01),whilep21WAFl/CIPlandcMycincreasedfrom(92.32±0.12)and(114.48±0.14)to(125.54±0.09)and(156.52±0.08)(P0.01).CONCLUSION:Thetimedependen

7、rotein.G2/Marrestisalsoassociatedwithapoptosis.[Keywords]Taoerqi;breastneoplasm;humanbreastcarcinomacelllinesMCF7;apoptosis【摘要】目的:研究桃兒七對人乳腺癌MCF7細(xì)胞增殖抑制和促進(jìn)凋亡的作用，探討其抗癌作用的機(jī)制.方法:應(yīng)用MTT法檢測桃兒七提取物對人乳腺癌MCF7細(xì)胞的生長抑制作用，倒置顯微鏡、HE染色及電鏡觀察細(xì)胞形態(tài)學(xué)改變,FCM分析細(xì)胞周期及凋亡率，免疫細(xì)胞化學(xué)法檢測細(xì)胞增殖凋亡調(diào)控相關(guān)蛋白的
8、表達(dá).結(jié)果淞兒七提取物可明顯抑制MCF7細(xì)胞的增殖，1,2,3mg/L桃兒七作用72h后，細(xì)胞生長抑制率分別為43.0%,54.9%,78.5%,抑制作用呈時(shí)間及濃度依賴性.形態(tài)學(xué)觀察到用藥后凋亡細(xì)胞典型的形態(tài)學(xué)特征，伴有多核細(xì)胞形成.FCM分析發(fā)現(xiàn)桃兒七阻斷MCF7細(xì)胞生長于細(xì)胞周期的G
