1、摘要摘要本試驗(yàn)收集了生產(chǎn)上栽培的5個(gè)白靈菇菌株,通過拮抗試驗(yàn)、酯酶同工酶分析及出菇試驗(yàn),對(duì)5個(gè)菌株進(jìn)行了生理生化特性和栽培特性分析。在親緣關(guān)系確定的基礎(chǔ)上,選取生產(chǎn)性狀優(yōu)劣互補(bǔ)的“中農(nóng)1號(hào)”和“中農(nóng)翅鮑”兩個(gè)菌株作為親本。出菇收集擔(dān)孢子,采用單孢分離技術(shù)分離兩個(gè)菌株的擔(dān)孢子,得到“中農(nóng)1號(hào)”單核體54個(gè),“中農(nóng)翅鮑”單核體41個(gè)。選取生長較快的兩親本單核菌絲進(jìn)行了雜交配對(duì),得到150個(gè)雜交組合菌株,根據(jù)鎖狀聯(lián)合有無進(jìn)一步確定融合子。采用拮抗、酯酶同工酶、原基誘導(dǎo)及出菇等方法對(duì)融合子進(jìn)行了進(jìn)一步鑒定和評(píng)價(jià)分析,結(jié)果表明:(1)通過拮抗、酯酶同工酶和出菇驗(yàn)證,5個(gè)白靈菇分
2、為兩大類群,一類以“中農(nóng)1號(hào)”為代表菌株;一類以“中農(nóng)翅鮑”為代表菌株。(2)通過鏡檢、拮抗、酯酶同工酶、原基誘導(dǎo)及出菇驗(yàn)證,150對(duì)雜交組合菌株中,99個(gè)菌株生長較快,有鎖狀聯(lián)合,75個(gè)與兩親本有明顯拮抗線,且具有兩親本菌株的典型酯酶同工酶條帶,確定是雜交菌株,其中50個(gè)雜交子具有出菇能力,出菇驗(yàn)證初步得到了3個(gè)優(yōu)良融合菌株。關(guān)鍵詞:白靈菇,單孢分離,雜交育種,篩選I萬方數(shù)據(jù)AbstractAbstractThephysiological,biochemicalandculturalcharacteristicsof5Pleurotuseryngiivar.tuol
