1、中草藥ChineseTraditionalandHerbalDrugs第39卷第12期2008年12月·1869··藥材與資源·巴戟天遺傳多樣性的RAPD研究X丁平,劉瑾,仰鐵錘,邱金英(廣州中醫(yī)藥大學(xué)中藥學(xué)院,廣東廣州510405)摘要:目的應(yīng)用隨機(jī)擴(kuò)增多態(tài)性DNA(randomamplifiedpolymorphicDNA,RAPD)標(biāo)記技術(shù)對(duì)廣東產(chǎn)5個(gè)類(lèi)群的栽培巴戟天進(jìn)行遺傳多樣性研究。方法應(yīng)用15種引物對(duì)64個(gè)個(gè)體進(jìn)行檢測(cè),應(yīng)用Popgen32軟件分析群體的遺傳多樣性。結(jié)果共得到224條帶,其中多態(tài)位點(diǎn)11
2、2個(gè),多態(tài)百分率為50100%。種群內(nèi)多態(tài)位點(diǎn)百分率在37105%~53113%,平均為45186%。Shannon指數(shù)和Nei指數(shù)均反映出巴戟天各類(lèi)群遺傳多樣性有一定的變化。Shannon指數(shù)為012876,各類(lèi)群的Shannon指數(shù)在011033~012362,Nei指數(shù)為011756,各類(lèi)群的Nei指數(shù)在010745~011540。結(jié)論目前栽培巴戟天居群間有一定的分化,并產(chǎn)生了多種農(nóng)家類(lèi)型,其中小葉巴戟天(POP2)遺傳多樣性明顯高于其他類(lèi)型,這些可能是導(dǎo)致目前栽培巴戟天質(zhì)量產(chǎn)生變異的主要原因。關(guān)鍵詞:巴戟天
