2、肝組織中擴(kuò)增出ApoE基因編碼序列,將該基因連接至pEGFP—c1載體上,利用雙酶切和測(cè)序?qū)χ亟M質(zhì)粒pEGFP.c1一ApoE進(jìn)行鑒定,并將重組質(zhì)粒pEGFP—c1一ApoE轉(zhuǎn)染PKl5細(xì)胞?!窘Y(jié)果】廣西巴馬小型豬ApoE基因編碼區(qū)序列長(zhǎng)954bp,編碼317個(gè)氨基酸,與GenBaIll【已公布的豬ApoE基因cDNA序列(NM214308)對(duì)應(yīng)區(qū)段同源性為100%。構(gòu)建的重組質(zhì)粒pEGFP.c1一ApoE轉(zhuǎn)染PKl5細(xì)胞48h后,在熒光顯微鏡下轉(zhuǎn)染細(xì)胞發(fā)出熒光,細(xì)胞形態(tài)清晰,可見(jiàn)細(xì)胞核?!窘Y(jié)論】廣西巴馬小型豬ApoE基因能與pEGFP-c1載體重組構(gòu)建pEGFP.
