1、2014年3J]色譜Vo1.32NO.3March20l4ChineseJournalofChromatography278~283研究論文DOI:10.3724/SP.J.1123.20l3.1l010摘要:應(yīng)川丁氣相色譜一質(zhì)譜聯(lián)J1](GC—MS)的代謝組學(xué)方法結(jié)合細(xì)胞用期實(shí)驗(yàn),研究制扇雕體外抑制人乳腺癌鄉(xiāng)【H胞MCF一7增殖的作用機(jī)理。代謝組學(xué)的研究結(jié)果表明:通過(guò)交偏最小方差判別分析(OPLS—DA)叮以很好地區(qū)分羽痢~Z雕f1:用的MCF一7細(xì)胞代謝譜與對(duì)照紺細(xì)胞代鰣譜,模型參數(shù)為:R=Ycum=0.988,QYcum=0.964VIP(variable
2、importanceintheprojection)值大于1的差異代謝物進(jìn)一步川t檢驗(yàn)進(jìn)干J-化分析,選擇t<0,05(VIP>1)的代f9If物作為羽扇豆醇作用組的生物標(biāo)志物,得到琥珀酸、磷酸、亮氨酸、亮氨酸等l1種代塒差異物。結(jié)合羽崩豆醇將細(xì)胞胤期抑制侄G1期這一現(xiàn)象,推測(cè)羽扇醇町能是主要抑制r三羧酸循環(huán)f}J的琥舶n先輔酶A的乍成和底物磷酸化,ij成ATP的反應(yīng)來(lái)抑制MCF一7細(xì)胞的增殖。本實(shí)驗(yàn)從代謝組學(xué)角度為乳腺螂抗jIf1瘸rdL$提供新的線索關(guān)鍵詞:氣卡l{色譜一質(zhì)譜;羽扇醇;MCF一7細(xì)胞;乳腺癌;抑制機(jī)理;交偏最小方差判別分析;細(xì)胞代謝組學(xué);細(xì)胞
