2、驗室——省部共建國家重實驗室培育基地,浙江省農(nóng)業(yè)科學(xué)院植物保護與微生物研究所,浙江杭州310021)摘要:對浙江省82個水稻立枯絲核菌菌株及13個標準融合群菌株5.8SrDNA—ITS(ITS)區(qū)間測序和分析,結(jié)果表明,ITS在標準融合群菌株間的差異遠大于AG1一IA融合群的水稻菌株間的差異,進化分析表明,ITS的差異可以有效甄別不同標準融合群菌株,但對AG1.IA融合群的水稻菌株區(qū)分能力非常有限。單倍型分析表明,這82個水稻菌株可以分為10個單倍型。田間菌株的單倍型多樣性(Hd)為0.2087,核苷酸多樣性為0.00033。其中單
3、倍型Hl為優(yōu)勢單倍型,占全部菌株的87.8%,而其他9個單倍型則各只有1株菌株。中性檢驗結(jié)果表明Tajima’SD測驗值為一2.26012(P<0.01),F(xiàn)u和Li’SD測驗值為一5.20827(P<0.02),暗示浙江省水稻紋枯菌菌株顯著偏離中性假說,在歷史上可能出現(xiàn)過種群擴張,并存在很強的選擇作用。關(guān)鍵詞:立枯絲核菌;水稻;轉(zhuǎn)錄間隔區(qū);序列測定;單倍型中圖分類號:Q78文獻標志碼:A文章編號:1004—1524(2014)03-0702-06DiversityresearchonRhizoctoniasolaniisolate
