1、HCN2陽性細胞在小鼠消化管肌間神經(jīng)叢內的分布熊承杰,梅峰,韓鵑,田衍平,江忠勇,周德山(第三軍醫(yī)大學組織學與胚胎學教研室,重慶400038)摘提要:目的探討(問題1:中文全稱?)(HCN2Asubtypeofchannelsofhyperpolarization-activatedcyclicnucleotide-gatedfamily,HCN2)HCN2刪除陽性細胞在小鼠胃腸道肌間神經(jīng)叢的分布。方法成年昆明小鼠的胃腸道,包括胃、小腸、結腸,制作全層鋪片免疫細胞化學染色HCN2陽性細胞和膽堿能神經(jīng)及cajal間質細胞(ICCs;interstitialCajalcells,ICCs)。結果成
2、年小鼠胃腸道肌間神經(jīng)叢的HCN2陽性細胞成簇緊密排列,多數(shù)陽性細胞為DogielIΙ型神經(jīng)元,陽性產(chǎn)物分別在細胞質胞漿和細胞膜,每個神經(jīng)節(jié)內可見數(shù)十個至數(shù)百個陽性細胞;神經(jīng)突起雖可見HCN2陽性產(chǎn)物,但主要存在于較粗的突起。HCN2陽性神經(jīng)元分布密度在胃腸道各段差異較大:結腸密度最高,胃次之,小腸比胃低,另外,;胃腸道的各截斷表達量也不一樣。利用膽堿能神經(jīng)元標志物(ChAT,膽堿乙酰轉移酶)和ICCs的標志物(c-kit蛋白)與HCN2進行雙重免疫染色可見HCN2與膽堿能神經(jīng)元共存,而未見HCN2與ICCs共存。結論HCN2離子通道廣泛表達于肌間神經(jīng)叢,存在于膽堿能神經(jīng)元,可能參與調控胃腸神經(jīng)

6、thosewhichwerethicker.ThedensityofdistributionforIRcellsforHCN2inthegastrointestinaltractisdifferent::colonishighest,stomachislowerthancolon,smallintestineislowest,andquantityofexpressionofeachcutofgastrointestinaltractisalsodifferent.ThemarkersofCholinergicnerves(ChAT)andICCs(c-kit)wereusedtodouble
7、-immunostainingwithHCN2,collocationofHCN2positivenervesandCholinergicnervescanbefound,butnocollocationofHCN2positivenervesandICCscanbefound.conclusionHCN2channelsarewidelydistributedinthemyentericnerv
