1、萬(wàn)方數(shù)據(jù)·322·生堡墅里叢壁苤查;!!!至±旦箜!!鲞箜!塑曼墅!』旦!旦!塑塑堡!£墅堡:壘2型!!!!!∑!!:!!!墮!:!體內(nèi)外人脂肪間充質(zhì)干細(xì)胞向肝樣細(xì)胞分化的實(shí)驗(yàn)研究殷莉波趙文秀尹震宇楊中萌張瓊王效民【摘要】目的分離、培養(yǎng)入脂肪問(wèn)充質(zhì)干細(xì)胞(hADsCs),研究其在體內(nèi)外向類肝樣細(xì)胞轉(zhuǎn)化的能力。方法采用I型膠原酶消化皮下脂肪,貼壁分選去除造血細(xì)胞的方法獲得較純的干細(xì)胞。通過(guò)對(duì)形態(tài)觀察、表面分子鑒定、透射電鏡內(nèi)部結(jié)構(gòu)掃描以及成脂、成骨定向誘導(dǎo)對(duì)脂肪間充質(zhì)干細(xì)胞進(jìn)行鑒定。用EGF、FG
2、F、OSM、HGF、TSA細(xì)胞因子體外誘導(dǎo)hADSCs向肝樣細(xì)胞分化。尾靜脈輸注hADSCs至急性肝損傷裸鼠,觀察hADSCs在裸鼠肝臟內(nèi)定植、向肝樣細(xì)胞分化情況,以及hADSCs對(duì)裸鼠肝損傷的保護(hù)作用。結(jié)果分離獲得的細(xì)胞呈長(zhǎng)梭形貼壁生長(zhǎng),低表達(dá)CD34、CD45,高表達(dá)CD73、CD90、CDl05,具有多向分化潛能。誘導(dǎo)細(xì)胞表現(xiàn)出肝樣細(xì)胞形態(tài),表達(dá)AFP、alb,并且和肝細(xì)胞一樣具有吸脂性。體內(nèi)誘導(dǎo):移植細(xì)胞在裸鼠體內(nèi)生存、分布,并且細(xì)胞表達(dá)人alb,細(xì)胞體積較裸鼠肝臟細(xì)胞大,形態(tài)不均一,與
