1、解剖學(xué)研究2015年第37卷第3期AnatRes,2015.V01.37.N0.3167·論著·神經(jīng)干細(xì)胞定向分化為多巴胺能神經(jīng)元過程中Mashl及Hesl基因的表達(dá)張君度,龍大宏,陳艷,劉菲菲,龍靜怡(廣州醫(yī)科大學(xué)人體解剖學(xué)教研室,廣東廣州510182)【摘要】目的建立神經(jīng)干細(xì)胞(NSCs)分化為多巴胺能神經(jīng)元的體外模型,并進(jìn)一步研究此分化過程中Mashl及Hesl基因的表達(dá)變化。方法無(wú)菌條件下取孕14—16d胎鼠端腦進(jìn)行NSCs體外培養(yǎng)。將NSCs進(jìn)行Nestin免疫熒光檢測(cè),對(duì)照組和實(shí)驗(yàn)組(GDNF+IL.1et聯(lián)合組)誘導(dǎo)2周后進(jìn)行B—tubulinⅢ和TH免疫熒
2、光染色,計(jì)算各組陽(yáng)性細(xì)胞率,熒光定量PCR技術(shù)檢測(cè)P4代NSCs分化1周和2周時(shí)Mash1及Hesl基因的表達(dá)變化。結(jié)果免疫熒光檢測(cè)結(jié)果顯示,NSCs的Nestin陽(yáng)性細(xì)胞率為(93.04+3.55)%,誘導(dǎo)分化2周后對(duì)照組、實(shí)驗(yàn)組p.tubulin111陽(yáng)性細(xì)胞率分別為(12.65+1.58)%和(33.95~2.97)%,兩組間比較差異存在統(tǒng)計(jì)學(xué)意義(P<0.05)。對(duì)照組、實(shí)驗(yàn)組TH陽(yáng)性細(xì)胞率分別為(1.46~0.41)%、(15.51~1.94)%,差異存在統(tǒng)計(jì)學(xué)意義(P<0.05)。熒光定量PCR結(jié)果顯示,NSCs誘導(dǎo)分化后,Mashl基因表達(dá)較未分化狀態(tài)時(shí)升高
3、,各組間比較差異存在統(tǒng)計(jì)學(xué)意義,實(shí)驗(yàn)組高于對(duì)照組(P<0.05),分化2周高于1周(P<0.01);Hesl基因于分化后表達(dá)降低,對(duì)照組與對(duì)實(shí)驗(yàn)組之間以及分化1周與2周之間比較差異沒有統(tǒng)計(jì)學(xué)意義(P>0.05)。結(jié)論GDNF和IL.1聯(lián)合作用能夠誘導(dǎo)NSCs定向分化為多巴胺能神經(jīng)元;Mashl與Hes1基因可能參與NSCs分化為多巴胺能神經(jīng)元的過程?!娟P(guān)鍵詞】神經(jīng)干細(xì)胞;分化;多巴胺能神經(jīng)元;Mashl;HeslTheexpressionofMashlandHeslduringinduceneuralstemcellsdiferentiatingintodopaminer
