1、摘要L一青霉胺是一種特效藥物的中問體,用于合成HIV一1蛋白酶抑制劑,具有很強(qiáng)的抗病毒能力,對艾滋病有明顯的預(yù)防和治療作用。從結(jié)構(gòu)上可以看出,L一青霉胺屬于氨基酸類化合物,含有一個(gè)活性碳原子。目前國內(nèi)僅有D一青霉胺的生產(chǎn)。本課題著力于縮短原有生產(chǎn)外消旋體青霉胺和L一青霉胺的工藝,主要研究以D一青霉胺為原料,乙酰氯為酰化劑,?;玫酵庀w乙酰D、L一青霉胺,產(chǎn)率為74.6%。乙酰一D、L一青霉胺在鹽酸中水解得到外消旋體D、L一青霉胺,產(chǎn)率為76.2%。采用優(yōu)先結(jié)晶法,逆向結(jié)晶法和生成非對映體鹽的方法來拆分D、L一青霉胺,但都沒
2、能拆分成功。本研究在采用L一(+)一氯霉胺為拆分劑,利用化學(xué)法直接拆分乙酰一D、L一青霉胺,獲得了成功。該法拆分效果好,乙酰一L一青霉胺的產(chǎn)率高,達(dá)到91.72%,拆分試劑L一(+)一氯霉胺的回收率為90.4%,并且拆分后得到的乙酰一L一青霉胺在鹽酸中水解制備出L一青霉胺,產(chǎn)率為71.4%,純度為97.34%。本文探討了溶劑量,化學(xué)試劑量,反應(yīng)溫度,反應(yīng)時(shí)間對乙酰一D、L一青霉胺產(chǎn)率的影響;同時(shí)探討了溫度與時(shí)間對D一青霉胺的消旋化進(jìn)程的影響,得出了消旋過程的動(dòng)力學(xué)方程;以及乙酰一L一青霉胺水解階段鹽酸的濃度,回流時(shí)間,PH值及
