2、stemcells,MSCs)在誘導(dǎo)條件下的成骨特性。方法:使用密度梯度法分離成年大鼠骨髓MSCs,以地塞米松、β-甘油磷酸鈉、維生素C為成骨誘導(dǎo)劑。結(jié)果:形態(tài)學(xué)表明,MSCs貼壁細(xì)胞呈集落生長,有成纖維細(xì)胞外觀。地塞米松低劑量組誘導(dǎo)7d后堿性磷酸酶(alkalinephosphatase,ALP)表達(dá)明顯升高,陽性細(xì)胞數(shù)為(15.1±2.6)個(gè),而對照組ALP表達(dá)較弱,陽性細(xì)胞數(shù)為(12.0±3.5)個(gè),兩組比較P<0.01。地塞米松成骨誘導(dǎo)劑低劑量組促進(jìn)MSC成骨,在誘導(dǎo)7d時(shí)即出現(xiàn)鈣化結(jié)節(jié),而對照組未出現(xiàn);地塞米松低劑量組誘導(dǎo)21d鈣化結(jié)節(jié)為(9.0±1.7
