2、中。通過(guò)三親接合將pBI2phyA質(zhì)粒由大腸桿菌導(dǎo)入根癌農(nóng)桿菌LBA4404菌株,獲得帶有phyA基因的根癌農(nóng)桿菌工程菌株LBA4404/pBI2phyA。用LBA4404/pBI2phyA轉(zhuǎn)化木薯外植體652個(gè),在含100mg/L卡那霉素和250mg/L氨芐青霉素的MS分化培養(yǎng)基上誘導(dǎo)分化芽,獲得24株抗卡那霉素轉(zhuǎn)化苗。以轉(zhuǎn)化苗和未轉(zhuǎn)化植株的總DNA為模板,用phyA基因的同源序列為引物,進(jìn)行聚合酶鏈?zhǔn)椒磻?yīng)(PCR),結(jié)果表明9株轉(zhuǎn)化苗的PCR結(jié)果為陽(yáng)性,初步證明目的基因phyA已經(jīng)轉(zhuǎn)入木薯中。關(guān)鍵詞:植酸酶基因;根癌農(nóng)桿菌;木薯;轉(zhuǎn)基因中圖分類號(hào):Q94312文獻(xiàn)標(biāo)識(shí)碼:APrel
